Growth hormone-releasing peptide 6 (GHRP-6) (developmental code name SKF-110679), also known as growth hormone-releasing hexapeptide, is one of several synthetic met-enkephalin analogues that include unnatural D-amino acids, were developed for their growth hormone-releasing activity and are called growth hormone secretagogues. They lack opioid activity but are potent stimulators of growth hormone (GH) release.
These secretagogues are distinct from growth hormone releasing hormone (GHRH) in that they share no sequencerelation and derive their function through activation of a completely different receptor. This receptor was originally called the growth hormone secretagogue receptor (GHSR), but due to subsequent discoveries, the hormoneghrelin is now considered the receptor's natural endogenous ligand, and it has been renamed as the ghrelin receptor. Therefore, these GHSR agonists act as synthetic ghrelin mimetics.
It has been discovered that when GHRP-6 and insulin are administered simultaneously, GH response to GHRP-6 is increased (1). However, the consumption of carbohydrates and/or dietary fats, around the administration window of GH secretagogues significantly blunts the GH release. A recent study in normal mice showed significant differences in body composition, muscle growth, glucose metabolism, memory and cardiac function in the mice being administered the GHRP-6 (2). There are still many questions regarding this fairly new compound, scientists are hoping to gain a better clinical understanding of the peptide through further research over the next few years.
GH secretagogues differ from exogenous rHGH in their effects primarily because endogenous GH contains all five isoforms of growth hormone, whereas exogenous GH contains only the 20 kilodalton isoform. Different isoforms affect tissues in discreet ways that the 20 kDa isoform cannot. Administration of GH secretagogues causes a pulse-release of GH from the pituitary which is cleared from the body within a few hours. This does not significantly raise plasma insulin-like growth factor 1 (IGF-1) levels.
GHRP-6 is a potent stimulator of natural Growth Hormone release. GHRP-6 is a Hexa-peptide that promotes food intake by stimulating hunger and helps increase energy metabolism. Growth Hormone Releasing Peptides, similar to GHRP-6, are most commonly used for treatment of Growth Hormone (GH) deficiencies, eating disorders, obesity, etc. Research has shown that use of these HGH Peptides increases lean muscle mass, strength, stamina and decreases body fat.
GHRPs are not simply surrogates of GHRH, instead GHRP-6 is an artificial activator of a separate newly discovered receptor called Growth Hormone Secretagogue Receptor (GHS-R). Soon Ghrelin was discovered, the endogenous ligand that binds to the GHS-R. Both Ghrelin and all the synthetic compounds such as GHRP-6 were termed "Growth Hormone Secretagogues" (GHSs). One side effect of GHRP-6 is a significant increase in appetite due to stimulating the release of Ghrelin, a peptide that is released naturally in the lining of the stomach that increases hunger and gastric emptying. Also, GHRP-6 causes stimulation of the anterior pituitary gland which causes an increase in Growth Hormone release. The increased amounts of Growth Hormone can cause the liver to secrete the hormone IGF-1, which improves the animal body's ability to burn fat and build muscle. Since GHRP-6 acts directly on the feedback loop which signals the inhibition of Growth Hormone release, GHRP-6 can re-stimulate the production of Growth Hormone.
How to use GHRP-6
GHRP-6 is most commonly provided in small vials of 5 mg, which should be stored under refrigeration. (It is acceptable however for them to be mailed unrefrigerated.) The vial is diluted with a convenient volume of sterile or bacteriostatic water. For example, the vial might be diluted with 2.5 mL of water, yielding a solution of 2 mg/mL (2000 mcg/mL.) After the water addition, the vial again will be stored under refrigeration.
When dosing, an appropriate volume will be drawn from the vial with (typically) an insulinsyringe, according to the desired dose and the concentration of the preparation. In the above example, a 100 mcg dose would require only 0.05 mL, or "5 IU" as marked on an insulin syringe. A 300 mcg would require 0.15 mL, or "15 IU" as marked on an insulin syringe.
Injection may be subcutaneous, intramuscular, or intravenous according to personal preference.
Dosing will ordinarily be at least twice per day and preferably 3x/day for best effect, taken at least 30-60 minutes before a meal and at a time of non-elevated blood sugar (in other words, after blood sugar has had time to fall since the most recent meal.) The amount taken generally will be from 50-300 mcg at a time. When using a GHRH along with GHRP-6, dosing should be reduced to 50-100 mcg at a time.
For increase in GH levels, higher doses within the suggested range definitely increase effect. With regard to healing benefit, for example for tendonitis, the low end of the range is often entirely sufficient and noticeably greater effect is not necessarily seen with increased dose.
While there is no sharp cut-off between a solution of GHRP-6 still being good and having lost potency with time, as a general guideline, a vial should be used within a month of having been reconstituted. Past this, I would discard the vial and start a new one.
